A MATE1 inhibition assay is an in vitro functional assay designed to determine the potential of a test compound to block the transport function of the Multidrug and Toxin Extrusion 1 (MATE1 or SLC47A1) transporter. The assay utilizes MATE1's role as a proton-cation antiporter to measure its active transport activity and the subsequent IC50 of the inhibitor. This is accomplished by intentionally reversing MATE1's physiological efflux direction to simplify detection.
