The P-gp inhibition assay is a functional in vitro study that quantifies the concentration-dependent disruption of the active efflux function of the human P-glycoprotein (P-gp or ABCB1) transporter. This ATP-driven efflux activity, which normally pushes drug substrates out of cells, is measured as a decrease in the P-gp-mediated transport of a probe substrate (like digoxin) when the test compound is present in a polarized cell monolayer or an inside-out membrane vesicle system.
