The MRP2 inhibition assay is a functional in vitro study that quantifies a compound's ability to block the active efflux function of the human Multidrug Resistance-associated Protein 2 (MRP2 or ABCC2) transporter. It typically uses inside-out membrane vesicles (IOVs) prepared from MRP2-expressing cells, which allows the transporter's ATP-dependent efflux activity to be measured as an easily quantifiable substrate accumulation into the vesicle.
