The MDCKII monolayer permeability assay utilizes a Madin-Darby canine kidney II (MDCKII) epithelial cell sub-line, which, when grown on porous transwell filters, rapidly differentiates to form a tight, polarized cellular barrier. This system is employed to measure the apparent permeability coefficient (Papp) of a compound across the barrier, providing a high-throughput screening tool to estimate passive transcellular permeability and the likelihood of intestinal absorption.
