The MDCK-BCRP permeability assay is a functional in vitro system used to quantify the overall directional permeability (Papp) of a compound across a cellular barrier. The assay utilizes a polarized cell monolayer (MDCK cells stably transfected with human BCRP) on a filter support, which allows for simultaneous measurement of both passive permeability (non-saturable movement across the lipid membrane) and BCRP-mediated active efflux (energy-dependent transport).
