Caco-2 permeability

definition

Caco-2 permeability assay is an in vitro ADME (Absorption, Distribution, Metabolism, and Excretion) assay that uses Caco-2 cell line (derived from human colon carcinoma) to assess bidirectional transport (apical to basolateral (A?B) and basolateral to apical (B?A)) across a confluent epithelial cell monolayer. Primarily used to predict in vivo absorption across the intestinal epithelium, this assay can also serve as a cost-effective early screen for passive permeability in drug discovery, including as a preliminary filter for compounds intended to cross other biological barriers such as the blood–brain barrier.
