Cell viability can be assessed by the presence of cytoplasmic esterases that cleave a lipid-soluble nonfluorescent probe to yield a fluorescent product. The product is charged and is thus retained within the cell if the membrane is intact. Typical probes include fluorescein diacetate (FDA), carboxyfluorescein, and calcein.
